Glenn Newman will be the TMO in the game, and Richard Kelly will be the Foul Play Review Officer (FPRO).
On the same day, in Paris, Paul Williams will referee his 40th Test when France hosts Wales in the opening game of the 2025 Six Nations.
O'Keeffe will also be an assistant referee when James Doleman referees the Scotland-Ireland game at Murrayfield on February 9. Richard Kelly will be the TMO.
Williams will be assistant referee to England's Matthew Carley when Italy hosts Wales in Rome on February 8. Newman will be the FPRO.
Australian referee Angus Gardner will also referee his 50th Test, a mark reached by 14 other referees when Ireland plays France in Dublin on March 8. He is the first Australian to achieve the milestone.
In the women's Six Nations, New Zealander Natarsha Ganley will referee Italy-Ireland at Parma on March 30, with Aaron Paterson as the TMO. Ganley will also be an assistant referee for England-Italy at York on March 23, with Paterson again as the TMO.
Fellow New Zealander Maggie Cogger-Orr will referee England-France at Twickenham on April 26.
